Cell alteration and aging as a process of the metabolism and tilted immune mechanisms

If the metabolism goes into the "red zone“ of excessive oxidative and nitrosative stress, alterations occur on membranes and within cells. To prevent the gene-programmed final surrender of apoptosis, damaged parts and organelles of the cells are being removed by autophagy before replacement with intact structures. To prevent these age-driving processes, it is crucial to re-balance the metabolism, both in regulative and nutritive ways. At the same time, the immune system watches, informs, reacts and induces change through cytokines. If this immune status equally enters the red zone, all effects are widely augmented.

The therapeutic strategy is to directly influence the metabolic pathways, to optimize the nutritive utilization, reduce or eliminate metabolic road blocks – and at the same time re-balance the immune response and its foundations.

Nutritional strategies to optimise autophagy/mitophagy

In a society where it is often the case of nutritional energetic surplus and poor nutrient rich intake, natural in house cleaning processes like autophagy may be severely reduced or highly unlikely. Nutritional strategies can in some aspects support this process whist maintaining health and correct nutritional support for our physiology. In this lecture I will be presenting applications of KD and TRF to optimise these aspects of metabolism including a personal research project and a scientific paper. I will cover the most common problems found in the application of nutritional strategies for KD and why we should estimate energetic intake differently in this context presenting a novel theory in energy estimation.

Applications of Fasting and Spermidine as Rejuvenation Strategies

One of the mechanistic aspects of rejuvenation through fasting is autophagy – a cellular “recycling” process that has been connected to many longevity methods so far. Hence, targeted activation of this cellular process could represent a viable strategy towards the
deceleration and possibly reversal of aging in general. Over the last two decades, fasting mimetics, many of which are autophagy activating substances, have been examined as possible longevity inducing agents.

One of the most prominent and emerging ones is spermidine – a polyamine that prolongs life and health span of various organisms in
autophagy-dependent manner. It is synthesised by our own body, but it call also be found in food. Recent clinical trials have shown evidence that an increased intake of spermidine in humans could deteriorate the effects of ageing. Hence, it could represent an effective strategy to reduce the rate or at least slow down the development of a range of age-related morbidities.

Mitophagy: New opportunities for nutrition in mitochondrial dysfunction

Mitochondria are not static but highly dynamic and constantly changing in shape, size and location within the cells. Dysfunction of mitochondria is associated with a wide-range of chronic diseases, with mitochondrial-targeted nutritional interventions becoming a well-established therapeutic approach. New discoveries related to ‘mitophagy’ are causing us to rethink the role of nutrition in mitochondrial dysfunction and improve treatment.

Removal of damaged mitochondria via autophagy (mitophagy) and the formation of new healthy mitochondria (biogenesis) is critical for cellular function and health. The balance between mitochondrial biogenesis and mitophagy determines the quantity and quality of mitochondria and is vital to adaptation to cellular stress and optimal metabolic function. Impaired mitophagy is emerging as a feature of chronic diseases, including cardiometabolic and neurodegenerative diseases.

Remarkably, several nutritional factors have recently been shown to influence mitophagy, suggesting an important opportunity for personalized nutritional interventions for the enhancement of mitochondrial function and metabolic health beyond traditional approaches. This talk covers recent advances in mitophagy and nutritional medicine, including the role of phytonutrients, phospholipids, metabolic endotoxemia, nutritional detoxification and intermittent fasting.
